CD74（又称为Major Histocompatibility Complex, Class II invariant chain）传统上被认为主要在抗原呈递细胞（APC）中表达，参与MHCⅡ类分子的折叠、运输和抗原呈递。有研究发现，在人肿瘤微环境（TME）中的调节性T细胞（Treg）表面，CD74被显著上调，成为一种肿瘤Treg的特异性标志物。 CD74在常规Treg中主要存在于胞内，但在肿瘤微环境中被诱导至细胞表面，成为肿瘤Treg的特异性标志物。它通过调控Treg的细胞骨架、迁移能力、细胞器稳态及转录活性，支持Treg在肿瘤内的积累和抑制功能。CD74的缺失会导致Treg在肿瘤中失能，促进肿瘤排斥，提示靶向CD74是一种有潜力的抗肿瘤策略。
