CXCR4是一种含七个跨膜结构域的G蛋白偶联受体（GPCR），是基质衍生因子-1（SDF-1/CXCL12）的特异性CXC趋化因子受体｡CXCR4最初被发现的身份是HIV入侵CD4+ T细胞的共受体之一，阻断CXCR4有望成为HIV治疗的新策略｡CXCR4信号通路在造血干细胞的迁移､增殖和静息及其在骨髓中的滞留中发挥重要作用，并介导B细胞CD20的上调｡CXCR4在23种以上的肿瘤中高表达（包括乳腺癌､卵巢癌､黑色素瘤和前列腺癌），而在正常组织中表达极低或不表达,其表达与肿瘤的存活､生长和向高SDF-1/CXCL12表达组织（如肺､肝､骨髓）的转移密切相关｡

图3. Immunohistochemistry Validation of CXCR4 in Human Spleen

Immunohistochemical analysis of paraffin-embedded human spleen tissue using anti-CXCR4 antibody (1009) at 5 ug/ml. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A Goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.
